The techniques commonly used for isolation of discrete colonies initially require that the number of organisms in the inoculums be reduced. Molten cooled agar (approx. 15 ml) is then poured into the petri dish containing the inoculum. Inoculum from a sample is pipette out and spread evenly on sterile nutrient agar by spreader.
